PBS Buffer Preparation pH 7.4 post we have briefly explained about 10X and 20X PBS buffer recipe and Tween-20 PBS buffer recipe preparation protocol.

PBS buffer recipe is widely used in biological research. It’s a water-based salt solution with Sodium chloride, Disodium phosphate, Monopotassium phosphate in some formulations. The buffer aids in the maintenance of a steady pH. The solutions’ osmolarity and ion concentrations are identical to those found in the human body.

Sodium chloride (NaCl)

Disodium phosphate (Na2HPO4)

Monopotassium phosphate (NaH2PO4).

Because it is isotonic and non-toxic to cells, PBS buffer recipe has a wide range of applications. It’s a liquid that can be used to dilute substances. It’s used to clean cell-filled containers.

PBS buffer recipe can be used as a diluent in ways to dry biomolecules because the water molecules inside it are organised around the substance to be ‘dried’ and adhered to a solid surface (for example, a protein). Denaturation and other conformational changes are prevented by the thin film of water that binds to the material.

PBS Buffer Preparation pH 7.4

10X PBS (0.1M PBS, pH 7.4)

Na2HPO4 (anhydrous): 10.9 g

NaH2PO4 (anhydrous): 3.2 g

NaCl: 90 g

Distilled water: 1000 ml

Mix to dissolve and adjust pH to 7.4. Store this solution at room temperature. Dilute 1:10 with distilled water before use and adjust pH if necessary for PBS buffer recipe 10X PBS (0.1M PBS, pH 7.4).

20X PBS (0.1M PBS, pH 7.4)

Na2HPO4 (anhydrous): 21.8 g

NaH2PO4 (anhydrous): 6.4 g

NaCl: 180 g

Distilled water: 1000 ml

Mix to dissolve and adjust pH to 7.4. Store this solution at room temperature. Dilute 1:10 with distilled water before use and adjust pH if necessary for PBS buffer recipe 20X PBS (0.1M PBS, pH 7.4).

10X PBS-Tween 20 (0.1M PBS, 0.5% Tween 20, pH 7.4)

Na2HPO4 (anhydrous): 10.9 g

NaH2PO4 (anhydrous): 3.2 g

NaCl: 90 g

Distilled water: 1000 ml

Mix to dissolve and adjust pH to 7.4 and then add 5 ml of Tween 20. Store this solution at room temperature. Dilute 1:10 with distilled water before use and adjust pH if necessary for PBS buffer recipe 10X PBS-Tween 20 (0.1M PBS, 0.5% Tween 20, pH 7.4).

20X PBS-Tween 20 (0.2M PBS, 1% Tween 20, pH 7.4)

Na2HPO4 (anhydrous): 21.8 g

NaH2PO4 (anhydrous): 6.4 g

NaCl: 180 g

Distilled water: 1000 ml

Mix to dissolve and adjust pH to 7.4 and then add 10 ml of Tween 20. Store this solution at room temperature. Dilute 1:10 with distilled water before use and adjust pH if necessary.
